$Space:: will not trigger if you hold shift and then hit space...

You want $+Space:: or $*Space::

WheelUp::a is logically equivalent to: Wheelup::Send {a down} Wheelup up::Send {a up} Mouse wheel does not have an up (Release) event like most keys do, so in effect, it is equivalent to Wheelup::Send {a down} , which would explain the behaviour you see. Rohwedder's solution should work around this.

Did you select "Block" for the keyboard input? That will probably stop the game from being able to see the keyboard inputs change state

Mbutton is for middle mouse, not Left Mouse. Just change MButton to LButton.
